Unit Introduction

SimuLab Laboratories Pty. Ltd. is a small laboratory and as a Laboratory Technician your responsibilities include the use of sophisticated chromatographic and electrophoretic equipment to perform analytical tests. In the following sections you will concentrate on the principles of operation and testing related to HPLC (high performance liquid chromatography), GC (gas chromatography) and SDS-PAGE (sodium dodecyl sulfate polyacrylamide gel electrophoresis).

You will also gain exposure to other techniques, specifically PC (paper chromatography), TLC (thin layer chromatography), affinity chromatography, gel filtration chromatography and western blotting.

Chromatography and electrophoresis are used to separate complex mixtures into their components and this process is used to identify and sometimes measure the levels of specified components in the samples. Your responsibilities extend to proper preparation to perform the work and finally to communicating the results to the client.

Note that the word ‘chromatography’ is commonly used to describe the family of chromatographic techniques. You will find that chromatography and its adjective – chromatographic - will be used frequently in this unit of study.
